Harnessing APCs and T Cells

Antigen-presenting cells (APCs) are the architects of the immune response by presenting antigens to T cells. Depending on how the antigen is presented, T cells can either become (a) pro-inflammatory “helper” CD4+ T cells or “killer” CD8+ T cells or (b) tolerogenic regulatory T cells (Tregs).

CD4+ T cells promote inflammaiton and help to active CD8+ T cells, which attack any cells presenting the corresponding antigens. Tregs, by contrast, reign in CD4+ and CD8+ T cells corresponding to specific antigens.


Antigen-Specific Immune Tolerance

BreezeBio’s lead programs induce immune tolerance. Non-immunogenic HNPs deliver mRNA encoding specific antigen(s) (with or without additional co-factors) to APCs. Presentation of antigens in the context of tolerance leads to unresponsive/anergic T cells and/or antigen-specific Tregs.

This approach has the potential to cure autoimmune diseases without broad immunosuppression, leaving the immune system otherwise intact to fight infections and malignancies.

In Vivo CAR-T

BreezeBio is leveraging our platform to re-engineer T cells directly in the body to generate reprogrammed CAR-T cells in vivo, which drives a stronger burst of efficacy for some forms of autoimmunity and cancer.

To treat autoimmune disease, in vivo CAR-T cells target B cells that produce autoantibodies. By transiently reducing the B cell population, the immune system is able to reset and restore homeostasis to spare healthy tissue.
